Why is it so big? Did you blow it up? I'd much rather see it at actual pixel size. Try to keep all the pixels the same size (the clouds look like they've been scaled up.)

In general, the pixels just look kind of sloppy. Some of the lines look like they need to be straightened up a bit. The quality of this could be improved a lot by just reading a couple of basic pixelling tutorials. (This one is a good start, and addresses some of the things I think you might not be familiar with, since you're new to pixel art.)

Other than that, though, you've got a cute style and some pretty decent color choices. I think you'd benefit a lot from trying something smaller - if you're used to working with vector graphics, you're probably used to working at whatever size you want and then resizing. With pixel art, you want to pick the size you want to work at and stick to it. If you want to work this big, I'd recommend splitting up the picture into smaller pieces so that you can focus on the pixels.

Mattam - That looks like it's for texturing polygons for 3d games. I don't think that rotating the tile is very plausible for these kind of tiles, since the view and the light source are not straight-down.

Already looking much better! What's that she's holding in her hand?

Personally I think she could use a little bit of outline, to set her apart from the background. For some of the lighter parts where you haven't shaded you, just sing the shadow color to shade might work. The shape of the face looks like it needs to be smoothed out a bit at one point, too.

Mattam's edit is a good suggestion. Honestly, I didn't even notice that some of the other stuff in there is quite nice because the floor was so overpowering.

What's up with the shadows halfway down the walls, though? (And on the top for that matter.) If they're straight walls, there shouldn't be any shading. Right now, it looks like the walls are bulging out. That's not too bad, it gives me a kind of sci-fi vibe, but when it's used on the doors as well, something seems off.

I really like all the different textures in the grass.

I think I liked the first sprite a lot better.

For a sprite like that, I'd say start with three (a midtone, a shade color, and the outline color) and add as you need them (like a highlight color, for instance.) If you want to do some anti-aliasing, you can add colors in between.

While choosing colors, keep in mind that you don't have to keep them on the same hue. You can get much more dynamic colors by moving toward blue/purple (as well as reducing the saturation) as your colors get darker. ;D

In any case, the best way to break up a grid to use different tiles. Don't just tile 1 tile, tile several with little changes. Don't forget to use some tiles that are just 1 color either.
Yes, I know. :3 I haven't made them yet, but I do plan to have a few of each tile. I want to get this one fixed first, though, so that I can make sure they all tile.

It's pillow-shaded. Don't shade in gradiations of a bunch of colors like that, it makes the shading blurry and indistinct. When you're shading, just make big areas of color (think like in cell-shaded anime.) After that, you can add more colors, but start with just a few.

There are some really good Touhou pixel art videos on YouTube if you want to get a look at how someone works while doing pixel art in the style you want.

The sprite is pretty cute, otherwise. I think the shading is your biggest problem right now. ;D Keep working at it! Hope I've helped~

Also, your aniamtion lacks weight. It doesn't feel like the walking cane is really being used to prop up the weight of your character.  
There's nothing to keep him standing if he lifts his cane and it's not going to work very well as an attack while moving either.

It could be that he isn't really using the cane to walk with, but rather just taking it with him to use as a weapon?

For some reason, I don't really like the arm swinging beside him in the walking animation. Also, he looks a bit like he's wearing war paint, hehe.

Your background is beautiful. ;D
