Fool, you've definitely pointed out a lot that I should change, thanks!  :)

I'm unsure of what to do with the hand that doesn't have the gun. It looks unnatural how I currently have it, but I don't want to change the pose to Fool's, for reasons that Sokota and Cure pointed out. Any opinions?

I didn't exactly understand what you just said. Could you show me by pointing an arrow at the place?

I added a bit more contrast, and did a bit of tweaking. I need to make the face displayed more prominently.

Well, you definitely need to make the scarf animated on the running pose.  ::)

Try copying the leg on the front, once you fix it, and shade it darker (Right now it's hard to tell the difference between the legs.) Then put it on behind the front leg.
Also, it looks like he is running slightly toward us (That's how it seems to me from looking at his face), but his hat looks a little too tilted in our direction.

It also seems like there's too much of a outline behind the ear on this animation:

Helm - You were right about the face. I was being a little lazy.

About your question, the background will be lighter and darker than the sprite, which will make the anti alias look bad.
