The whole scale issue might just be the way you laid out the tiles on the screen. if you were to make something with bigger spaces, it would probably look alright.
the character does look bigger than what I usually see, but it depends on the game-play I suppose

OK I made an edit, using your colors (I think I may have changed the jeans a bit, but it would be the same) and you can see how I made bigger shapes, and less texture on the vest and legs.
I think your sprite is fairly small to add detail such as wrinkles, so you'd be better off just doing the main volumes

First thing that springs to mind is how big the character is. I feel he's a bit big for the platforms. other than that, maybe the vertical tiles look very straight and neat. but its solid work.

my suggestion then is to reduce the contrast between mid tone and highlight. that will help reduce the glossy effect

is the hood and vest ment to be glossy? cause right now they look a bit glossy and noisy

I think in such small sizes, dithering can be hard to make look good. maybe you could instead try and use the transition to create some texture rather than dithering
The chalice could be wider, more room for detail. right now its looks like a flute glass for champagne

what you call "oblique 90" I usually call "top-down"

I think considering the outside view had this slight depth with the houses, you could do the oblique 45. It would fall closer in terms of style

on the references you posted initially, there's almost no shading done on the dress, and the volume is suggested buy seam lines or by the outline occasionally curving around the body shapes.
maybe you could try that and see where that leads. if you feel comfortable understanding how the edge curves work, you could then gradually add shades on the inside of the shape that match the body's volume.

I edited your animation. All I did was look at the characters eye from one frame to another, and made sure he didn't move too much.
there was one frame I editeded (frame 6 I think) because the face was slightly 3/4 view, and it was the only frame so it looked odd.
