Looks quite good!

A few points for improves, maybe:
The mouth might be darker, it's not obvious at all in a casual glance at your av.
Dithering has the opposite effect of what you wanted: breaks down the head round shape into flat planes. You need more single-pixel-level control there.
It generally could use more contrast, I think.
And, those blue eyes?  ???

I hadn't noticed those drains!

Are they really that close to the houses and pillars?
Is there no railing or protection?

I'm so nerdish about drains, sewers and that stuff.

Still starving for contrast.

The hats shadow is almost invisible, why not use the darkest brown there?
And you need one more highlight color on the face: nose, glasses.

Clouds are not far from good.
I'm not sure they need dithering, but if they do, reduce it and break it in really very small patches. As soon as you have 3 checkerboard lines, you end up with the opposite of what you tried to achieve: a new flat obvious cluster.
And I'm sure the car needs no dithering! Shiny curved surface will have neat linear reflections, you might try that.

I like the colors, the cropping too. H<3K could use some AA (that grey might work fine) but you're probably not there yet.

You need to check the metal/glass transition: I like simple line as on the left side, but it gets lost on the more realistic other side with the heavy dark line.

Not too late I hope!

Boring??? Well, how about some background now, maybe with that bird in it?
Or some illustration of this:

This has come some way, congrats!

Those tiles are beautiful! The tree too.

The vines may not be so great, compared to the rest. I can't really point why, but they look a little cheap.

What are those doors (those below the diaphragms) for? To be pasted upon any other tile? Nothing wrong with them anyway.

I'm inexperienced at spriting, but I'd suggest to remove the outline first. At this small size, you need every pixel and color for shading and volume.

i made sure that the light was closer to the right than the left.

Far from enough! Just by 1px in fact, it's almost un-noticeable.
Also, the hat (mostly) and glasses (maybe) should cast a shadow.

Shame on me for not having searched wikipedia.  :(

Nice patterns, but there are strong readability issues.

Mirroring tiles destroys any hint at a light source you ever had, if any, so the patterns appear flat.
The only place with a little depth from light is the 2 rightmost dark tiles.
You will have to shade your tiles independently: choose a light source, decide whether the patterns stick out or recess, and stick to it.

I'd have some tiles with less detail too, and arrange them more randomly.

Having the same patterns on fore- and background is a good idea, but doesn't help readability, too. I'd reduce bg contrast.

Same thing with the sprite: he's got similar colors and values as the bg, so he blends into it. He's also 'pillowshaded', which makes him blend even more.
Again, think of a light source, and keep him contrasted, lots.

In this low-saturation setting, the green gun looks a bit weird.

Got one, not pixel specific though.

@ Ben2theEdge: what is negative space?
(I'm asking you specifically because from the search I did, it looks you actually mention it the most, but I'll gladly read anybody else's views too!)

I've been reminded of it about a piece in my PJ gallery (sorry to link to my own work, just convenient here) (wink @ ska!).
http://www.pixeljoint.com/pixelart/48570.htm

I feel I got the point about composition in that piece (have I?), but the general notion is eluding me.

Is there more to it than "a nice empty space for good composition"?
