What Panda says is right, your piece cannot be considered in any way as plain pixel art, as it's done using line and filling tools, without any visible pixel per pixel refinement.
The advice I could give is, as pixel art is a very difficult exercise, you could try making the same piece in a smaller pic, say up to 128x128
It will let you concentrate more on details and pixel work.
What do you think about it ?

what do you mean by add substance? you mean like clothes, more fleshy not to blok-ish?
