After you beat the boss of Downpour II, a boarded-up door appears. It takes two bombs to open. This leads to the Mines (or Ashpit).

With the knife in tow, play through the game normally until you hit Depths II (or Dank Depths/Necropolis). Kill the Mom boss.

Do not go into the Womb.

Instead, look for a weird, red, fleshy door in the starting room of the floor or the boss room. Walk up to it. The knife you spent the last 30 minutes assembling will fly out of your hands and stab the door. It swings open.

Inside is a boss fight against Mom’s Heart. It’s harder than the standard version, but if you’ve made it this far, you can handle it. Once the heart dies, the room fills with blue light and a hole opens in the floor.

Welcome to the Corpse.
